The role of the ubiquitin-proteasome pathway in rhTRIM5α restriction of HIV-1
نویسندگان
چکیده
Methods First, rhTRIM5a was examined by immunofluorescence and characterized with specific ubiquitin antibodies. Next, mutations were made in rhTRIM5a to prevent involvement in the ubiquitin-proteasome pathway, and mutants were analyzed by immunofluorescence, flow cytometry and real-time PCR. A mutant virus was constructed to prevent ubiquitination of capsid, and used to infect cells expressing rhTRIM5a to characterize restriction and reverse transcription. Finally, a fluorescently tagged subunit of the proteasome, LMP2-GFP, was coexpressed with rhTRIM5a and analyzed by fixed and live cell microscopy.

Suppressor of Cytokine Signaling 1 Counteracts Rhesus Macaque TRIM5α-Induced Inhibition of Human Immunodeficiency Virus Type-1 Production
Old world monkey TRIM5α is a host factor that restricts human immunodeficiency virus type-1 (HIV-1) infection. Previously, we reported that rhesus macaque TRIM5α (RhTRIM5α) restricts HIV-1 production by inducing degradation of precursor Gag. Since suppressor of cytokine signaling 1 (SOCS1) is known to enhance HIV-1 production by rescuing Gag from lysosomal degradation, we examined if SOCS1 is i...
متن کاملHijacking of the Ubiquitin/Proteasome Pathway by the HIV Auxiliary Proteins
The ubiquitin-proteasome system (UPS) ensures regulation of the protein pool in the cell by ubiquitination of proteins followed by their degradation by the proteasome. It plays a central role in the cell under normal physiological conditions as well as during viral infections. On the one hand, the UPS can be used by the cell to degrade viral proteins, thereby restricting the viral infection. On...
متن کاملAdaptation of HIV-1 to rhTrim5α-mediated restriction in vitro.
Recently, gene therapy with rhTrim5α, an innate restriction factor which blocks HIV-1 at a post entry step, have been shown to be applicable as treatment in vitro. However, HIV-1 might adapt to replicate in the presence of rhTrim5α due to its high mutation rate. Here we observed that two different HIV-1 isolates were able to replicate in cells expressing high levels of rhTrim5α. Escape mutation...
متن کاملExpression Status of UBE2Q2 in Colorectal Primary Tumors and Cell Lines
Background: Activation of the ubiquitin-proteasome pathway in various malignancies, including colorectal cancer, is established. This pathway mediates the degradation of damaged proteins and regulates growth and stress response. The novel human gene, UBE2Q2, with a putative ubiquitin-conjugating enzyme activity, is reported to be overexpressed in some malignancies. We sought to investigate the ...
متن کاملAssisted Evolution Enables HIV-1 to Overcome a High TRIM5α-Imposed Genetic Barrier to Rhesus Macaque Tropism
Diversification of antiretroviral factors during host evolution has erected formidable barriers to cross-species retrovirus transmission. This phenomenon likely protects humans from infection by many modern retroviruses, but it has also impaired the development of primate models of HIV-1 infection. Indeed, rhesus macaques are resistant to HIV-1, in part due to restriction imposed by the TRIM5α ...
